Problem
An enzyme E binds a substrate S reversibly to form a complex ES, which then converts irreversibly to product P and releases free enzyme:E+Sk−1⇌k1ESk2E+PEnzyme is present at a fixed total concentration [E]0=[E]+[ES], and substrate is in large enough excess that [S] can be treated as constant during the early, initial-rate period over which [ES] has reached a quasi-steady state, d[ES]/dt≈0.
((a)) Write the rate equation for [ES] (formation minus loss), impose the steady-state condition, and eliminate [E] in favour of [E]0, [ES] and [S]. Show that the initial rate v=d[P]/dt=k2[ES] takes the Michaelis-Menten form v=KM+[S]kcat[E]0[S], and give kcat and KM in terms of k1, k−1 and k2.
((b)) For an enzyme with k1=2.0×107M−1s−1, k−1=300s−1, k2=100s−1 and [E]0=1.0×10−8M, compute KM and the initial rate v at [S]=5.0×10−5M.
((c)) Compute the dissociation constant Kd=k−1/k1 of the ES complex for this enzyme, compare it with KM from part (b), state the condition on k2 relative to k−1 under which KM→Kd, and explain why that condition does not hold here.
